What Is Munapsys™? Origin and Design
Munapsys™ was designed through an approach known as in silico design, which uses computational algorithms to simulate interactions between molecules and target proteins. The peptide was optimized to compete with the Munc-18 protein, a key component of the SNARE complex, interfering with the release of acetylcholine, which is responsible for muscle contraction.
It is made up of 99.4% naturally derived ingredients according to the ISO 16128 standard. It comes as a highly soluble aqueous solution and is used at concentrations ranging from 2% to 5%.
How It Works
Pre-synaptic action: acetylcholine release
In the neuromuscular system, contraction is activated by the release of acetylcholine (ACh) from the synaptic vesicles of the motor neuron. This release depends on the correct assembly of the SNARE protein complex, made up of Munc-18, syntaxin, SNAP-25, and VAMP.
Munapsys™ is designed to interfere with the Munc-18 protein, reducing its ability to bind to syntaxin and thereby hindering the formation of the SNARE complex. The result is a lower release of acetylcholine, producing an attenuated, temporary, and reversible effect.
Post-synaptic action: the muscular response
The second characteristic of Munapsys™ concerns the muscles. Unlike Argireline® (Acetyl Hexapeptide-8), which acts only at the level of the nerve signal, this peptide also modulates the expression of genes involved in muscle contraction.
In experimental models using human myocytes, the manufacturer reports a reduction in intracellular calcium mobilization, myosin levels, and the expression of certain contractile genes (ATP2A, TNNC1, MYH1, ACTA1, CACNB1, SCN3A).
This dual level of action — pre- and post-synaptic — is what distinguishes Munapsys™ from other muscle-relaxing peptides on the market.
Data Reported by the Manufacturer
The testing portfolio includes in vitro studies on neuronal and muscle cell models, ex vivo studies on neuron-myofiber co-cultures, and in vivo studies on a group of female volunteers aged between 35 and 55.
The in vitro tests report a reduction in the Munc-18/syntaxin interaction, a decrease in exocytosis in neurons, and lower myosin levels in treated myocytes.
Regarding the clinical test, conducted for 14 days on 18 female volunteers using a 3% cream and evaluated with PRIMOS and VISIA instruments, the manufacturer reports a reduction in skin roughness and the depth of expression lines, with results observable as early as the first week.
As with all patented cosmetic actives, these data come from tests conducted by the ingredient manufacturer and not from independent clinical studies published in peer-reviewed journals. The in vivo test sample was also small (18 subjects) and the duration was short: these findings indicate potential efficacy in a formulation, not definitive proof. The mechanism of action involving the SNARE complex, however, is described in the scientific literature.
Which Wrinkles Does It Target?
Munapsys™ delivers the best results on dynamic wrinkles, which are caused by the repeated movement of the facial muscles:
- glabellar lines, between the eyebrows;
- forehead lines;
- crow’s feet;
- perioral lines.
Its effect is more limited on static wrinkles already structured in the dermis due to the loss of collagen and elastin. In that case, active ingredients that work on the dermal matrix are needed, such as signal peptides or collagen stimulators.
Safety and Tolerability
According to the safety data provided by the manufacturer, Munapsys™ has a zero skin and eye irritation profile in in vitro tests on reconstructed epidermis, no sensitizing potential in the HRIPT, and good stability in formulations. It can therefore also be used on sensitive skin.
