Acetyl Tetrapeptide-5, marketed under the name Eyeseryl®, is one of the few cosmetic active ingredients developed specifically to address a precise concern: puffy under-eye bags.

What Is Acetyl Tetrapeptide-5?

It is a bioactive peptide, meaning a molecule capable of influencing specific biological processes. Peptides are chains of amino acids and form the building blocks of structural proteins such as collagen, elastin, and keratin.

In cosmetics, peptides are classified into three categories according to their mechanism:

  1. Signal peptides: stimulate the synthesis of collagen and elastin and slow their degradation;
  2. Neurotransmitter-inhibiting peptides: reduce the contraction of facial muscles, helping smooth expression lines;
  3. Enzyme-inhibiting peptides: block the activity of specific enzymes. Acetyl Tetrapeptide-5 belongs to this category.

How Does Acetyl Tetrapeptide-5 Act on Under-Eye Bags?

The main mechanism is the inhibition of ACE (Angiotensin-Converting Enzyme), the enzyme that regulates fluid pressure in blood vessels and capillary permeability.

When capillary permeability increases, fluids pass more easily into the surrounding tissues. In the under-eye area—where the skin is thin and lymphatic drainage is naturally slow—this results in classic eyelid edema: morning puffiness that tends to decrease throughout the day.

By reducing vascular permeability and promoting drainage, Acetyl Tetrapeptide-5 acts on the cause of fluid retention rather than the symptom.

What Does the Data Say?

Tests conducted by the manufacturer, Lipotec, report a visible reduction in the appearance of under-eye bags after approximately 28 days of application, with improvements observed in some subjects as early as two weeks, along with increased skin elasticity in the eye area.

As with most cosmetic active ingredients, these data come from tests conducted by the company that produces the ingredient, not from independent clinical studies published in peer-reviewed journals. This is standard industry practice, but it is important to know: they indicate potential efficacy, not independent clinical evidence.

The mechanism of action, however—the inhibition of ACE and its effect on vascular permeability—is supported by established pharmacological principles.

Why Does the Eye Area Need Dedicated Active Ingredients?

The eye area has characteristics that make it unique:

  • the skin is much thinner than elsewhere on the face, making blood vessels more visible;
  • collagen and elastin break down more quickly;
  • the stratum corneum is thinner, with a reduced barrier function;
  • lymphatic drainage is less efficient, which promotes fluid accumulation.

The manufacturer’s data indicate approximately 28 days of consistent application, with some improvement observable as early as two weeks. As with all peptides, the effect depends on regular use: when application is discontinued, the condition tends to return to its previous state.

Does It Also Work on Dark Circles?

It mainly targets dark circles of vascular origin, the bluish type associated with fluid retention. For pigmented dark circles, which are more brownish, brightening active ingredients such as niacinamide and vitamin C are needed.
